<p>Mitochondrial dynamics govern cardiomyocyte fate under hypoxic stress, yet the relative contributions of DRP1-driven GTPase hydrolysis and receptor-scaffolded fission through MFF/FIS1 complexes remain incompletely resolved. Using primary neonatal rat ventricular cardiomyocytes subjected to controlled hypoxia–reoxygenation, we systematically compared pharmacological DRP1 inhibition (Mdivi-1), siRNA-mediated MFF/FIS1 knockdown, and combinatorial suppression across indices of mitochondrial morphology, membrane potential (ΔΨm), cytochrome c release, and caspase-3 activation. Super-resolution STED microscopy and flow cytometric analyses revealed that MFF/FIS1 scaffolding predominantly initiates pathological fission events, whereas DRP1 GTPase activity amplifies downstream apoptotic signaling. Combinatorial inhibition achieved 74% reduction in apoptotic index compared to single-pathway targeting. These findings reframe the hierarchical architecture of the mitochondrial fission cascade and suggest dual-pathway therapeutic targeting as a superior cardioprotective strategy.</p>Skyler WalkerAdrian ScottRobin Miller

<p>Mitochondrial cristae architecture is a critical determinant of oxidative phosphorylation efficiency and apoptotic regulation. This study examines hypoxia-driven OPA1 oligomerization and its downstream effects on cristae junction width, respiratory supercomplex assembly, and cytochrome c sequestration in HeLa cervical carcinoma cells. Using a combination of focused ion beam scanning electron microscopy (FIB-SEM), proximity ligation assay (PLA), and Blue Native PAGE, we quantified ultrastructural remodeling events under 1% O₂ conditions over 6–48 hours. Our data demonstrate that sustained hypoxia induces a statistically significant narrowing of cristae junctions (p < 0.001) concurrent with elevated OPA1 high-molecular-weight oligomer formation and enhanced OXPHOS supercomplex I+III₂+IV stoichiometry. These findings establish a mechanistic link between inner mitochondrial membrane fusion dynamics and metabolic adaptation in hypoxic tumor microenvironments, with implications for mitochondria-targeted cancer therapeutics.</p>Jordan LopezRiley SmithSkyler Phillips
